I need some advice on where to go and how long it takes between each destination!

I'll be living in Ecuador from April to August next year. From August 1 to 25 I have free reign of the continent, and want to see as much as possible. I want to get a rough idea ahead of time so i can book a flight in one shot and save some money. I'm thinking somehow getting from quito to lima (bus? how long), spending sometime seeing peruvian sites, and then hopping a plane to either santiago or buenos aires, traveling between those two cities, and then heading back to NYC from whichever one i end up in. any suggestions? are there places far from the cities that i MUST see, and how long does it take to get to them? Obviously im a bit limited on time to see so many countries, so i have to pick and choose a little bit. i just dont want to miss anything amazing (yeah, right) or waste time just because i booked my flight out of the wrong city!

i did a route like that, here are the bus times
heading north:
Quito - Otavalo 3 hours (a must see, beautiful indigenous craft markets)
then heading south:
quito - baños 3 hours (baños is good for outdoor activities)
baños - riobamba 1 hour (indigenous food markets on saturdays)
riobamba - guayaquil 6 hours
guayaquil - montañita 4 hours (surfers beach)
guayaquil - mancora 9 hours (surfers beach)
mancora - chiclayo 5 hours (good city wuth nice historical sights &museums)
chiclayo - trujillo 3 hours (more historical sights)
trujillo - lima 8 hours
