Busing around South America

How do the bus tickets and stops work?

A friend and I are planning on traveling around South America starting in Rio and doing a big U shape and ending up in Quito. We are thinking that we will have to probably throw in a couple of internal flights just so h=that we can cover the ground but the rest of the time we are thinking of busing it.

Can someone help and tell me whether you are able to buy a bus ticket and hop on and off the bus to break up the journey, rather than having to do a 12 hour journey, or do you have to buy separate tickets and if so does this work out more expensive?

You will need to buy separate tickets, but in general they are very good value, so even if breaking a journey costs a little more, it's still affordable. It's worth booking first class buses for longer journeys, and on these you will normally book your seat in advance (hence you couldn't reuse a ticket for the same destination by breaking the journey as someone else will prob have your seat).
