Spain:
The best way to move around Spain is by car. Try renting one.
For Southern Spain check out Crown Car Hire:

Ottherwise better by bus than by train.

Barcelona is a must. I recommend you Seville and Granada (including the Sierra Nevada-Alpujarras area). San Sebastian and Santiago are really nice but far. If you can afford it, try driving around Mallorca Island, in spring it is a little paradise on earth. An interesting route along the southwest coast is Malaga-Marbella-Gibraltar-Tarifa-Conil-Cadiz-Jerez (gougeous view of Africa and the best beaches in Spain but the water's still cold in May) and the inner of Cadiz province.
...
From England to everywhere else: Ryanair

-London Stansted to Granada, Seville, Jerez and others-
From France, Italy to Spain: Vueling

Around Spain (and Balearic Islands): Air Europa, Spanair
